What is Tesamorelin?

Tesamorelin is a stabilized analog of the 44 residue growth hormone releasing hormone. As Egrifta it is FDA approved for one indication: reducing excess abdominal fat in adults with HIV associated lipodystrophy. That approval rests on phase 3 trials. Research grade tesamorelin is not the approved drug product and has no other approved use.

Synonyms
TH9507; Egrifta (approved drug product)
Classification
Synthetic analog of full length growth hormone releasing hormone (GHRH 1-44)

What is Tesamorelin?

Tesamorelin adds a trans-3-hexenoyl group to the N terminus of GHRH(1-44), which protects it from rapid enzymatic breakdown.

Research context

Phase 3 trials in HIV associated lipodystrophy were published from 2007, with later studies of liver fat and fat quality in people with HIV.

Mechanisms being investigated

  • GHRH receptor activation, increasing pulsatile growth hormone release and downstream IGF-1.

Research status

Approval status: FDA approved (limited indication). Highest evidence tier in the cited literature: clinical trials.

FDA approved (Egrifta) for reduction of excess abdominal fat in HIV associated lipodystrophy. Not approved for any other use.

Established findings

  • Efficacy on visceral fat in HIV associated lipodystrophy from randomized trials.
  • Its regulatory approval for that single indication.

Preliminary or experimental findings

  • Effects in other populations, including cognition studies, which are investigational.

Testing considerations

  • Large 44 residue peptide: high resolution MS and a well resolved HPLC method are needed to separate closely related impurities.

General methods are explained in purity versus identity testing.

What is still unknown

  • Long term effects outside the approved population.

Research use limitations

This page summarizes published research and regulatory status. It is not medical advice and does not describe any use in people. Material supplied by ENOS Labs USA is for in-vitro laboratory research only and is not a drug product, even where a related drug has been approved.
